Molekulare Analyse von Physcomitrella knock out Mutanten

Projektbeschreibung:
Das Moos Physcomitrella patens ist bisher die einzige Landpflanze, bei der fremde DNA nicht willkürlich ins Genom eingebaut wird, sondern gezieltes Gentargeting über homologe Rekombination möglich ist. In der ersten Projektphase (1999-2002) wurden zunächst eine EST-Datenbank mit mehr als 100.000 ESTs erstellt und über PEG-vermittelte Protoplastentransformation 75.000 Physcomitrella-Mutanten produziert. Alle Transformanten wurden physiologisch und phänotypisch charaktierisiert und dann kryokonserviert. Im Folgeprojekt (2003-2005) erfolgte zunächst die metabolische Analyse von rund 10.000 Physcomitrella-Mutanten. Diese Analysen waren Grundlage für die Auswahl von Pflanzen mit kommerziell interessanten phänotpyischen Veränderungen (erhöhter Vitamingehalt, veränderte Aminosäurezusammensetzung u.ä.). Die molekularbiologische Analyse der ausgewählten Transformanten führte zur Aufdeckung neuer Genfunktionen in Physcomitrella patens.
